STI Swab & Urine Kit is designed for a rapid and efficient purification of high quality bacterial, parasite or viral DNA from human swabs (such as buc- cal, throat, nasal, pharyngeal, penile, prepuce, vaginal, and rectal swabs) as well as from urine. The isolation protocol and buffer formulations were optimi- zed for high isolation efficiency and DNA purity.
Description
DNA purification procedure utilizes spin mini columns with membranes which efficiently and selectively bind nucleic acids. A swab or urine sample is subjected to enzymatic lysis by Proteinase K in SU-L Buffer. In this step, cell walls, membranes and proteins are degraded by lysis buffer and Proteinase K. After addition of chaotropic salts, lysate is applied to purification mini column membrane and DNA is bound. A three-step washing stage effectively removes impurities and enzyme inhibitors. The purified DNA is eluted using either a low ionic strength buffer (SU-E Buffer), distilled water or TE Buffer. The purified DNA can be used in PCR and real-time PCR.
